Lyophilization removes water by freezing a material and then lowering pressure so ice changes directly to vapor. The process relies on sublimation, the phase transition from solid to gas without an intermediate liquid state. Because the material remains frozen during primary drying, the structure often stays porous. This porous matrix can rehydrate quickly when water is added back. The low pressure also allows vapor to leave the solid matrix without boiling.
A typical cycle begins with freezing, which fixes the material into a solid and determines ice crystal size. Primary drying then raises heat under vacuum so ice sublimes, often near or below the collapse temperature of the formulation. Secondary drying removes bound water that remains after ice is gone, usually by gently warming the product. Each stage balances heat input against pressure to avoid melting or structural damage. Temperature probes and pressure sensors guide the transition between stages.
In practice, lyophilization is slower and more energy intensive than simple drying. Cycle times can range from hours to several days depending on load, container, and formulation. Amorphous materials may require excipients that help preserve structure during freezing and drying. The method is widely used for biological materials, pharmaceuticals, and foods where heat drying would cause unacceptable change. Open questions remain about scaling cycles between laboratory and production equipment, and this gap affects technology transfer.
Quality control for lyophilized products focuses on appearance, moisture level, reconstitution time, and structural integrity. A cake should be uniform, intact, and free of meltback or collapse. Moisture level is measured by Karl Fischer titration or thermogravimetric analysis. Reconstitution time reflects pore structure and formulation. Visual inspection and vial integrity checks detect cracks, stopper defects, or particulate matter. These checks are often performed on samples from each batch. They help confirm that the drying cycle performed as intended.
Storage stability depends on water content, oxygen exposure, and temperature. Lyophilized solids are hygroscopic and can absorb water if exposed to humid air. Vials are usually sealed under vacuum or inert gas with rubber stoppers and aluminum crimps. Storage temperatures range from room temperature to refrigerated or frozen conditions depending on the material. Stability studies track potency, moisture, and physical form over time. Accelerated conditions can reveal sensitivity but may not predict long-term behavior.

Lyophilization, also called freeze-drying, is a dehydration process in which a solvent, usually water, is frozen and then removed by sublimation under reduced pressure. The method preserves heat-sensitive materials that would degrade in conventional drying. Large-scale use grew during the mid-twentieth century for blood plasma and antibiotics, and it later expanded to vaccines, enzymes, foods, and advanced materials. The process produces a dry, porous solid that usually reconstitutes rapidly. It is distinct from simple evaporation because the solvent bypasses the liquid phase during primary removal.
The process generally proceeds in three stages: freezing, primary drying, and secondary drying. During freezing, controlled cooling converts water into ice and may also crystallize or vitrify solutes. In primary drying, the pressure is lowered below the triple point, and heat is supplied so ice sublimes directly to vapor. Secondary drying removes water that remains bound to the solid matrix, yielding a low final water content. Product temperature must stay below the collapse or glass transition temperature to maintain structure. Cycle design therefore balances shelf temperature, chamber pressure, and time.
After lyophilization, a product's quality depends on residual moisture, cake appearance, and reconstitution time. Residual moisture is often measured by Karl Fischer titration or thermogravimetric analysis. A low moisture content can slow chemical degradation, but overly dry cakes may be brittle or slow to dissolve. Stability studies track these attributes over months under defined temperature and humidity conditions. Batch records link these measurements to specific process runs and help identify trends before a product fails specification.
Storage conditions for dried products usually aim to exclude moisture and oxygen. Vials are sealed under vacuum or with an inert gas, and stoppers must maintain a barrier during transport. Temperature recommendations vary; some materials remain stable at room temperature, while others need refrigeration or frozen storage. Humidity control is critical because dried cakes can absorb water rapidly once a container is opened. Desiccant packs and moisture-barrier bags add further protection during shipping.
Formulation composition influences whether freeze-drying produces an intact cake or a collapsed mass. Excipients such as sugars and polymers can raise the collapse temperature and provide bulk during drying. The critical temperature for primary drying is often the collapse temperature or the glass transition temperature of the maximally concentrated phase. If the product temperature exceeds this threshold, the frozen matrix may soften and lose structure. Established practice therefore links shelf temperature and chamber pressure to the formulation's thermal properties.
The physics of freeze-drying couples heat transfer, mass transfer, and phase change. Heat supplied through the shelf must reach the sublimation front without melting the ice or degrading the product. Water vapor then travels through the already dried layer and leaves the chamber, where low pressure and cold traps keep it from returning. The dried layer acts as a resistance to vapor flow, so drying rate changes as the front recedes. Open questions remain about how pore structure and formulation heterogeneity affect drying uniformity at larger scales.
== History == The PUN was founded in 1957 as a political platform to support the presidential candidacy of "Papa Doc" Duvalier in the 1957 general election. In the aftermath of the July 1958 coup attempt, the Tonton Macoute — officially named Volunteers of the National Security (French: Volontaires de la Sécurité Nationale, VSN) — was formed as the paramilitary wing of the PUN. In 1963, other political parties in Haiti were outlawed, making the PUN the single political party of the nation. When "Papa Doc" Duvalier died in 1971, his son "Baby Doc" Duvalier succeeded him as the President of the country and the leader of the party. The party would remain in power until the fall and exile of "Baby Doc" Duvalier in 1986, at the height of the Anti-Duvalier protest movement, after which the party would enter a low profile stage, including the change of its name to National Progressive Party (French: Parti progressiste national, PPN). After the return of "Baby Doc" Duvalier from exile in 2011, the party began a process of reconstitution under its original name from 2014, and started opening departmental offices. Suffering a setback with the death of "Baby Doc" Duvalier in 2014, the party nominated Marc-Arthur Drouillard as its candidate in the 2015 presidential election. Towards that time, the legal representative of the party was Philomène Exe.
Mated females then seek a suitable place to begin a colony. There, they break off their wings using their tibial spurs and begin to lay and care for eggs. The females can selectively fertilise future eggs with the sperm stored to produce diploid workers or lay unfertilized haploid eggs to produce drones. The first workers to hatch, known as nanitics, are weaker and smaller than later workers but they begin to serve the colony immediately. They enlarge the nest, forage for food, and care for the other eggs. Species that have multiple queens may have a queen leaving the nest along with some workers to found a colony at a new site, a process akin to swarming in honeybees.
Four days after obtaining the "authority of a sovereign state" in 1955, West Germany joined NATO. The UK and the USA retained an especially strong presence in West Germany, acting as a deterrent in case of a Soviet invasion. In 1976, West Germany became one of the founding nations of the Group of Six (G6). In 1973, West Germany—home to roughly 1.26% of the world's population—featured the world's fourth-highest GDP of 814,796 million compared to East Germany's 129,969 million, together combining for 944,755 million and accounting for 5.9% of the world total. In 1987, the FRG held a 7.4% share of total world production.

Relaxin family peptide hormones in humans are represented by seven members: three relaxin-like (RLN) and four insulin-like (INSL) peptides: RLN1, RLN2, RNL3, INSL3, INSL4, INSL5, INSL6. This subdivision into two classes (RLN and INSL) is based primarily on early findings, and does not reflect the evolutionary origins or physiological differences between peptides. For example, it is known that the genes coding for RLN3 and INSL5 arose from one ancestral gene, and INSL3 shares origin with RLN2 and its multiple duplicates: RLN1, INSL4, INSL6.
Preclinical testing consists of animal pharmacology and toxicology studies to assess whether the drug is safe for testing in humans. Also included are any previous experience with the drug in humans (often foreign use). Manufacturing Information includes composition, manufacturer, and stability of, and the controls used for, manufacturing the drug. Used to ensure that the company can adequately produce and supply consistent batches of the drug. Investigator information on the qualifications of clinical investigators, that is, the professionals (generally physicians) who oversee the administration of the experimental drug to the study subjects. Used to assess whether the investigators are qualified to fulfill their clinical trial duties. Clinical trial protocols are the centerpiece of the IND. Detailed protocols for proposed clinical studies to assess whether the initial-phase trials will expose the subjects to unnecessary risks. Other commitments are commitments to obtain informed consent from the research subjects, to obtain a review of the study by an institutional review board (IRB), and to adhere to the investigational new drug regulations. An IND application must also include an Investigator's Brochure intended to educate the trial investigators of the significant facts about the trial drug they need to know to conduct their clinical trial with the least hazard to the subjects or patients. Once an IND application is submitted, the FDA has 30 days to object to the IND or it automatically becomes effective and clinical trials may begin.
=== Mutagens === One of the most common base analogs is 5-bromouracil (5BU), the abnormal base found in the mutagenic nucleotide analog BrdU. When a nucleotide containing 5-bromouracil is incorporated into the DNA, it is most likely to pair with adenine; however, it can spontaneously shift into another isomer which pairs with a different nucleobase, guanine. If this happens during DNA replication, a guanine will be inserted as the opposite base analog, and in the next DNA replication, that guanine will pair with a cytosine. This results in a change in one base pair of DNA, specifically a transition mutation. Additionally, nitrous acid (HNO2) is a potent mutagen that acts on replicating and non-replicating DNA. It can cause deamination of the amino groups of adenine, guanine and cytosine. Adenine is deaminated to hypoxanthine, which base pairs to cytosine instead of thymine. Cytosine is deaminated to uracil, which base pairs with adenine instead of guanine. Deamination of guanine is not mutagenic. Nitrous acid-induced mutations also are induced to mutate back to wild-type.

According to Reuters, the conflict involving Iran has resulted in increased oil prices, elevated inflation rates, and a subsequent rise in the cost of living within the United States. Furthermore, a majority of voters in 2025 report experiencing a decline in their economic well-being under the administration of President Trump.
The British historian Richard Overy wrote the huge demands of the Four Year Plan "...could not be fully met by a policy of import substitution and industrial rationalisation", thus leading Hitler to decide in November 1937 that to stay ahead in the arms race with the other powers that Germany had to seize Czechoslovakia in the near-future. At the Hossbach conference on 5 November 1937, Hitler announced that seizing Czechoslovakia would increase the supply of food under German control, which in turn would lessen the need to import food, thereby freeing up more foreign exchange to import raw materials necessary for the Four Year Plan's targets. The Hossbach conference was largely taken up with an extended discussion about the necessity of bringing areas adjunct to Germany under German economic control, by force if necessary, as Hitler argued that this was the best way to win the arms race. Hitler stated: "areas producing raw materials can be more usefully sought in Europe, in immediate proximity to the Reich". Overy wrote about Hitler's attitude to the Reich's economic problems that: "He simply saw war instrumentally, as the Japanese had done in Manchuria, as a way to expand the German resource base and to secure it against other powers". At the time, Czechoslovakia had Europe's 7th largest economy, and easily the most modern, developed, and industrialized economy in Eastern Europe.
This caused another rapid expansion, particularly in Germany. Between 1877 and 1887, 130 German patents for azo dyes were filed and 105 new dyes made it to market. It also lead to a difference in how chemical companies interacted with consumers. German dye firms developed in-house marketing and distribution capabilities coordinated directly with their research and development departments. Paul Schützenberger, in response to what he had seen at the 1878 Universal Exposition commented, "The abundance, the variety of combinations is such that we do not know whether to be more amazed by their multiplicity or by the imagination required to name them. Indeed, it is by the thousands that dyers create, every season, new colors for their sample cards." Professional societies based on the synthetic dye industries began to form. By the First World War, the largest number of dyes sold in the market fell into the class of azo dyes. 1885, an azo-naphthol, Para-red, became the first water-insoluble organic pigment not containing acidic or basic groups.

Primary drying removes ice by sublimation under vacuum. Secondary drying removes water that is bound to the material, often by warming the product after most ice has left. Both stages occur below temperatures that would cause unwanted melting.
Sublimation requires the solvent to remain solid so vapor leaves without passing through a liquid phase. If the product melts, the porous structure can collapse and drying becomes uneven. Maintaining frozen conditions preserves the intended physical form.
No, freeze-drying is a dehydration method, not a sterilization step. It can reduce water activity and limit microbial growth during storage, but it does not reliably kill microbes or remove endotoxins. Sterility must come from separate validated processes.
Karl Fischer titration is a common method, using coulometric or volumetric detection. Thermogravimetric analysis can also measure weight loss on heating. Results depend on sample handling because the dried solid can absorb moisture quickly.
